Abstract

Probiotic microorganisms have been increasingly applied in the medical and food industries due to their health benefits. This study aimed to construct and optimize a multiplex PCR kit for identifying simultaneously three probiotic strains Lactobacillus acidophilus, Streptococcus thermophilus and Bacillus subtilis. By using three specific primers slpA, lacS and sboA respectively, the multiplex PCR had been designed and integrated for the single reaction. Based on these findings, a multiplex PCR kit was amplified DNA products respectively the three bacteria strains without cross-reactivity. The optimal PCR condition including 75 µM MgCl₂, 300 µM dNTPs and priming temperature in 61 °C was explored and designed. This multiplex PCR kit had been applied in 10 commercial probiotic products with limit of detection (LOD) in demonstrating high sensitivity at 10/10 samples of practical test. This method indicated a high sensitivity, specificity and applicability. It also offered an effective tool for quality control and verification of probiotic components in commercial products.
