QUANTIFICATION OF TOTAL POLYPHENOL CONTENT IN FRUITS AND BULBS OF Allium schoenoprasum L. USING UV-VIS SPECTROSCOPY WITH FOLIN-CIOCALTEU REAGENT

This study was conducted to develop a UV-Vis spectrophotometric method using the Folin-Ciocalteu reagent for the rapid and accurate quantification of total polyphenol content in the fruits and bulbs of Allium schoenoprasum L. The optimal conditions for the complexation reaction were determined, including a pH range of 8.67–8.95 and a molar ratio of Na2CO3/Folin of 3.6, corresponding to an initial Folin reagent concentration of 0.20 mol/L (10% Folin reagent) and a Na2CO3 concentration of 1.2 mol/L, with a reaction time of 2 h at room temperature. The proposed method demonstrated high sensitivity, with MDLS (US.EPA) = 0.049 mg/L; LOD (AOAC) = 0.051 mg/L, LOQ = 0.155 mg/L, and exhibited a wide linear range (LOL) of 0.2–20 mg/L, making it suitable for determining polyphenol content in plant samples with high polyphenol concentrations while reducing errors associated with sample dilution. The study also established the conditions for polyphenol extraction from the fruits and bulbs of Allium schoenoprasum L., using 70% ethanol for the bulbs and 50–70% ethanol for the fruits. Extraction was performed using ultrasonic treatment at 30 °C for 30 min or immersion in a thermostatic bath at 70 °C for 30 min. The results revealed that the polyphenol content in the bulbs of Allium schoenoprasum L. was higher than that in the fruits, at 9.21 ± 0.29 mg/g dry weight (or 2.48 ± 0.08 mg/g fresh bulb), compared with 7.97 ± 0.26 mg/g dry weight (or 2.38 ± 0.08 mg/g fresh fruit), respectively.
