EXTRACTION OF TOTAL POLYPHENOLS FROM PERSIMMON PEEL USING DEEP EUTECTIC SOLVENTS

Polyphenols are naturally occurring bioactive compounds renowned for their strong antioxidant properties, making them highly valuable in the food, pharmaceutical, and cosmetic industries. In this study, we investigated the efficiency of deep eutectic solvents (DESs) for extracting polyphenols from persimmon peel (Diospyros kaki L.), aiming to enhance both yield and antioxidant activity while promoting a greener alternative to traditional solvents. Our findings identified choline chloride/lactic acid (ChCl/LA) with 30% water content as the optimal DES formulation. Under the optimized conditions, a material-to-solvent ratio of 1:40 (w/v) and an extraction time of 120 minutes, the total polyphenol content and antioxidant capacity, assessed through DPPH and ABTS assays, were significantly enhanced. Compared to extraction with 70% ethanol, polyphenol yield increased by 6.8 times, while antioxidant activity was 1.6 and 4.3 times higher for DPPH and ABTS, respectively. Microscopic analysis of DES-treated persimmon peel revealed the presence of structural modifications, such as the formation of cracks and pores, which likely facilitated solvent penetration and improved mass transfer efficiency. These results underscore the potential of DESs as a sustainable and efficient alternative for polyphenol extraction, offering an eco-friendly solution for valorizing plant-based bioactive compounds.
