NGHIÊN CỨU BIẾN TÍNH POLYSACCHARIDE TỪ RONG Ceratophyllum submersum BẰNG PHƯƠNG PHÁP CARBOXYMETHYL HÓA

Polysaccharide là nhóm hợp chất sinh học cao phân tử có nhiều hoạt tính sinh học và được chú ý trong nhiều lĩnh vực nghiên cứu nhờ tiềm năng ứng dụng như y sinh và dược phẩm. Ceratophyllum submersum là loài rong nước ngọt chứa hàm lượng polysaccharide đáng kể. Tuy nhiên, các nghiên cứu về biến tính cấu trúc để cải thiện hoạt tính sinh học của polysaccharide từ loài rong này vẫn còn khá ít. Nghiên cứu này được thực hiện với mục tiêu tìm ra điều kiện tối ưu cho quá trình biến tính polysaccharide bằng phương pháp carboxymethyl hóa và đánh giá ảnh hưởng của quá trình này đến hoạt tính sinh học. Nội dung khảo sát liên quan đến điều kiện carboxymethyl hóa bao gồm nồng độ acid chloroacetic (MCA), nhiệt độ phản ứng và thời gian phản ứng. Kết quả cho thấy điều kiện tối ưu đạt được tại nồng độ MCA 2,5 M, nhiệt độ 60 °C và thời gian phản ứng 4 giờ, cho mức độ thay thế DS tương ứng là 0,876 ± 0,032; 1,134 ± 0,063 và 1,313 ± 0,038. Đánh giá sự biến đổi cấu trúc bằng phổ FT-IR cho thấy sự xuất hiện của nhóm CH2COO- thay thế nhóm OH- vào mạch carbon. Hoạt tính sinh học được đánh giá thông qua khả năng kháng oxy hóa bằng các phương pháp 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), nitric oxide (NO), khử sắt (III) thành sắt (II) (RP) và khả năng kháng khuẩn. Polysaccharide sau biến tính thể hiện khả năng bắt gốc tự do mạnh hơn với giá trị IC50 lần lượt là 181,65 µg/mL (ABTS), 63,79 µg/mL (NO), tính khử sắt thể hiện qua sự tăng độ hấp thụ, sau khi polysaccharide biến tính giá trị độ hấp thụ cao hơn so với khi chưa biến tính, đồng thời có khả năng ức chế Salmonella và Staphylococcus aureus. Kết quả nghiên cứu cho thấy việc biến tính cấu trúc polysaccharide bằng phương pháp carboxymethyl giúp cải thiện đáng kể hoạt tính sinh học, qua đó khẳng định tiềm năng ứng dụng của polysaccharide từ Ceratophyllum submersum trong các lĩnh vực y học, dược phẩm và thực phẩm chức năng.
